As early as 1927-1937, Professor Raper H. S. of Manchester, England, published many papers in the Journal of Biochemistry. Through both in vitro and in vivo experiments, it was proved that the melanin in hair was tyrosine, which was catalyzed by human biological enzymes, and generated 5,6-dihydroxyindole (DHI) through a series of biochemical reactions. DHI was the precursor of melanin, which was oxidized into melanin to make the hair appear naturally black. When human hair follicle cells cannot produce enough DHI, hair will gradually turn white due to a lack of melanin. Melanin not only gives hair fibers a natural color, but also interacts with sunlight to prevent radiation damage to the inner layer of hair, thus playing a role in protecting hair quality. Inspired by this, scientists have been seeking to generate DHI in vitro and simulate the formation of natural melanin in hair, in order to achieve true healthy natural black hair technology. However, due to the great difficulty of this melanin biomimetic technology, there have been basically no real melanin black hair products approved for market in the past century.
Pure 5,6-dihydroxyindole (DHI) is a colorless needle shaped crystal with a melting point of 140 ℃. It is soluble in hot water and insoluble in petroleum ether; It is stable in a crystalline state, but quickly oxidizes into substances similar to melanin in a slightly alkaline solution. Due to its low irritation to human skin, it is used in some washing products to replace commonly used aniline compounds (which have carcinogenic, teratogenic, and allergic adverse effects) as a black active ingredient. For example, Meiyuan's hair dye in Japan uses it as one of the main active ingredients. It is also a good antioxidant and can be used as an intermediate in the synthesis of some amino acids, alkaloids, and tryptamines. In recent years, with the development of new varieties of fine chemicals, the use of 5,6-dihydroxyindole has also been constantly emerging. Many units have conducted synthesis research on this product, but there have been no reports of mass supply.
